W No More Heroes III wcielamy się ponownie w ekscentrycznego profesjonalnego zabójcę o imieniu Travis Touchdown. Bohater po dziesięciu latach powraca do fikcyjnego kalifornijskiego miasta Santa Destroy, które było areną jego przygód w poprzednich tytułach z serii. Na miejscu odkrywa, że nad metropolią lewituje ogromna platforma z kosmitami na pokładzie. Okazuje się, że przewodzi im Jess Baptiste VI znany jako FU. Jego celem jest zniszczenie Ziemi wraz z dziewiątką swoich pobratymców. Jedynym człowiekiem, który decyduje się mu postawić, jest właśnie Travis Touchdown. Nasz bohater uzbraja się w swoją specjalną energetyczną katanę oraz rękawicę „Death Glove” i wyrusza na pozornie niemożliwą misję uratowania planety.
W trakcie rozgrywki Goichi Suda próbuje wielokrotnie zaskoczyć gracza brutalnością, wulgarnością oraz abstrakcyjnością tego, co dzieje się na ekranie. No More Heroes III to spektakl przemocy w komiksowym opakowaniu, pełen nawiązań do popkultury.
